36.Kovac\'s reagent is used in Indole test. Kovacs reagent is 4 (p)-dimethylamino benzaldehyde.
Indole test is used to determine the ability of an organism to split indole ring from tryptophan.
Tryptophan is hydrolysed by tryptophanase to produce indole. Indole production is detected by
Kovac’s reagent with the formation of red color in the culture broth.
37. Oxidase. Oxidase test is done to detect the presence of cytochrome oxidase presence.
N,N,N,N-tetramethyl-p-phenylenediamine (TMPD) is the reagent used. Purple color production
indicates positive result.
