Bacterial Community Composition in the Growth Process of Pleurotus eryngii and Growth-Promoting Abilities of Isolated Bacteria(Original Article) RAFIA ANJUM ID:2022764

Introduction Pleurotus eryngii is an edible mushroom. Bacterial community helps to improve nutrition uptake, promote the growth of host fungi , prevent pathogen contamination. Aim of the Research: The aim of this study is to investigate the bacterial community in different growth stages(Vegetative and Reproductive growth phase) of Pleurotus eryngii by high-throughput sequencing technology to explore the relationship between interacting bacteria and the growth and development of Pleurotus eryngii

Materials and Methods Sample collection: Samples were collected from different growth stages. It was mixed and frozen by liquid nitrogen. 2.DNA Extraction: DNA was extracted by Easy pure Genomic kit . 16s rDNA was amplified targeting V3 and V4 region. 3. High –Throughput Sequencing: Amplifications were sequenced by Illumina Hiseq and OTUs were assigned into taxonomic categories . 4. Cell Free Fermentation Broth Assay: The growth speed was analyzed by adding cell free fermentation broth.

Result Analysis Figure :1 Chord diagram visualizing relative abundances of the dominant bacterial phyla and genera in different growth stages of P. eryngii . The ten most abundant phyla and the genera with relative abundances of > 1% were showed.

Result Analysis Figure 2:Mycelia growth rate of P. eryngii cultured in media with and without the different volumes of B. cereus Bac1 fermentation broth.

Discussion: Ten abundant phyla with 14 abundant genera being shared across all growth periods. The results showed that when a 5 mL cell free fermentation broth was used, the growth speed of P. eryngii hyphae was enhanced.
