COLORIMETERY PRINCIPLE PROCEDURE APPLICATION

C O L O R I M E T E R Y Colorimetry is the field of determining the concentration of a coloured compound in a solution . A colorimetery ,also known as a filtre photometer ,is an analytical machine that act as the tool quantity a solution concentration by measuring the absorbance of a specific wavelength of light .

COLORIMETER A colorimeter is a device used in colorimetery that measures the absorbance of particular wavelength of light by a specific solution .

Colorimeter device is mainly used in industries and laboratories for analyzing the color quality of the products along with color measurements . Colorimeter is very sensitive device that can measure the concentration and intensity of a particular color that is used in a product.

There are two types of colorimeters: Color desitometer A densitometer is a device that measures the degree of darkness (the optical density) of a photographic or semitransparent material or of a reflecting surface. Color photometer A spectrophotometer (color  spectro ) is a color measurement tool for measuring color .

PRINCIPLE OF COLORIMETER The intensity of color is directly propotional to the concentration of the compound being measured. Colorimetery is the technique frequently used in biochemical investigations ,involves quantitative estimation of colors .

Color can be produced by any substances when it binds with color forming chromogens . The difference in color intensity results in the difference in the absorption of light .

WORKING OF PRINCIPLE OF COLORIMETER Working of colorimeter is mainly based on the BEER LAMBERT’S LAW. This law states that the light absorption when passes through a medium are directly propotional to the concentration medium When a colorimeter is used there is a ray of light with a certain wavelength is directed towards as solution.

Before reaching solution the ray passes through a series of different lenses. These lenses are used for navigation of colored light in colorimeter. Colorimeter analyses the reflected light and compares with a predetermined standard . Then a microprocessor installed in the device is used for calculation of absorbance of the light by the solution.

If absorption of solution is higher then there will be more light absorbed by the solution If the concentration of the solution is low then more lights will be transmitted through the solution.

LAMBERT’S LAW Lambert’s law states that the amount of light transmitted decrease with increase in the thickness of layer of colored solution .

According to beer’s law when light passes through a colored solution amount of light transmitted decreases with increase in concentration of colored substance .

COMPONENTS OF COLORIMETER Light source Adjustment slit Considering lens Set of coloured filters Cuvette ( sample chamber to hold working solution) Detector to measure the transmitted light Meter to display the output from the detector

LIGHT SOURCE Generally a tungsten or xenon lamp is used to produce light.

FILTER It is made up of colored glass and is used to choose the light of narrow wavwlength . CUVETTE Used to hold the solution of sample. DETECTOR Used to detect the light transmitted through the sample.

PROCEDURE IN COLORIMETRY In colorimetric estimations, it is necessary to prepare a test a standard and a blank Test solution is prepared by treating a specific volume of the specimen with reagents . Filtre is inserted in photometer. Cuvette is filled with water about three fourth and placed in position. Light is allowed to pass through the cuvette absorbance is adjusted to zero .

Blank solution is taken in another cuvette and is placed in the cuvette compartment. Absorbance is noted(B) Absorbance of test(T) Standard(S) Satisfactory results obtained when ,T&S are in the range of 0.1-0.7

APPLICATION OF COLORIMETERS Food ingredients Textile products Beverages Chemical solution and many others.

USES OF COLORIMETER Used in hospitalsto estimate biochemical samples such as Urine‘ Cerebrospinal fluid Plasma Serum etc..

Used in manufacturing of paints . Used in textile and food industry Used to test water quality . Used to determine the concentration of haemoglobin in the blood .
