Virology

Definition
•A virus is a tiny, infectious particle that can
reproduce and replicate only inside living cells.
•A virus composed of a DNA or RNA genome, but
not both inside a protein shell called a capsid.
Some viruses have an external membrane called
envelope.
•Viruses are not considered living. They're very
small, much smaller than the cells of living things,
and are basically just packages of nucleic acid and
protein.

• some viruses also have an external lipid
membrane known as an envelope, which
surrounds the entire capsid, this called
enveloped virus.
•Non enveloped (naked) viruses are highly
dangerous than enveloped because it’s resist
to detergent and some disinfectant.

•All Herpesviruses are structurally similar:
•Enveloped (the envelope contains viral
glycoprotein spikes and Fc receptors)
•Icosahedral symmetry

Modes of transmission
•1.across the placenta,
•2.breast milk,
•3.via saliva in young children,
•4.in adults can be transmitted sexually,
•5.blood transfusion,
•6.organ transplants.

Diagnosis:
•1) PCR assay is now superior to virus isolation because
the virus is slow growing making the diagnosis late, so
PCR detection of replicating virus and viral load assays
are now very useful in diagnosing CMV infection.
•2) Monoclonal Ab against viral Ag can be used in
detecting virus-positive leukocytes.
•3) Culturing in special tubes called shell vials coupled
with the use of immunoflourecent antibody Diagnosis
can be made within 72 hours.
•4) Staining of owl's eye inclusion bodies (intranuclear).
•5) four fold rise in the Ab titter is also diagnostic.

•Treatment:
•Ganciclovir can be used in treatment of
CMV retinitis and pneumonia in patients
with AIDS.
•Control:
•Both live and recombinant
cytomegalovirus vaccines are under
development.
