Dexona injections, manufactured by Zydus
Cadila Healthcare Ltd., India is a synthetic
corticosteroid exhibiting both anti-
inflammatory and immuno-suppressant
properties. This medicine is used for
replacement therapy in secondary adrenal
insufficiency arising from insufficient
corticotrophin secretion.

It is also indicated for allergic disorders such as
bronchial asthma and allergic skin reactions, blood
disorders such as leukaemia, thrombocytopoenia and
haemolytic anaemias, selected collagen and
rheumatic disorders (only rarely in rheumatoid
arthritis), gastrointestinal disorders such as
inflammatory bowel disease, ulcerative colitis,
connective tissue disorders such as arteritis, systemic
lupus erythematosus (but not scleroderma), some skin
diseases such as pemphigus, oedema, some eye
disorders, certain neoplastic disorders such as cerebral
neoplasm, Cerebral oedema (raised intracranial
pressure), auto-immune, endocrine, pulmonary,
secondary hypercalcaemia, and acute leukaemia in
children.

➢The initial dosage of Dexamethasone Sodium
Phosphate Injection varies from 0.5 mg to 9 mg
a day depending on the indication being
treated. In less severe diseases doses lower
than 0.5 mg may suffice, while in severe
diseases doses higher than 9 mg may be
required.
➢Acute Allergic Disorders: Dexamethasone
sodium phosphate injection, USP 4 mg/mL;
first day, 1 or 2 mL (4 or 8 mg), intramuscularly.

➢Cerebral Edema: Dexona injection (Dexamethasone
Sodium Phosphate Injection) is generally administered
initially in a dosage of 10 mg intravenously followed by four
mg every six hours intramuscularly until the symptoms of
cerebral edema subside.
➢Intra-Articular, Intralesional and Soft Tissue Injection: Intra-
articular, intralesional and soft tissue injections are usually
used when affected joints or areas are limited to one or two
sites. Dosage and frequency of injection varies depending on
the condition and the site of injection. The usual dose is from
0.2 mg to 6 mg. The frequency usually ranges from once
every three to five days to once every two to three weeks.

Generic Dexamethasone Injections
(Dexona) have to be stored at controlled
room temperature i.e. from 20°C to 25°C
(68°F to 77°F). Protect from light.
Sensitive to heat. Protect from freezing.
Keep this as well as all other medicines
away from children and pets.

Precautions is necessary when corticosteroids are used
in patients with the following conditions and frequent
monitoring is necessary: Hypertension, Hypothyroidism,
Congestive Heart failure or recent myocardial infarction,
Liver failure, Renal insufficiency, Diabetes mellitus or in
those with a family history of diabetes, Osteoporosis,
Glaucoma, Patients with a history of severe affective
disorders particularly of steroid induced psychoses,
Epilepsy and/or seizure disorder Peptic ulceration,
Previous steroid myopathy, Tuberculosis, patients with
myasthenia gravis receiving anticholinesterase
therapy since corticosteroid use may decrease plasma
anticholinesterase activity, Patients with Cushing's
disease and Patients with thromboembolic disorders.

Corticosteroids may exacerbate systemic fungal
infections and, therefore, should not be used in the
presence of such infections unless they are needed to
control drug reactions due to amphotericin B.
This medicine has to be administered with caution in
patients with Duchenne’s muscular dystrophy since
transient rhabdomyolysis and myoblobinuria have
been reported following strenuous physical activity.
Administration of live virus vaccines, including
smallpox, is contraindicated in individuals receiving
immunosuppressive doses of corticosteroids.
Patients who are on medicines that suppress the
immune system are more susceptible to infections than
healthy individuals. In such children or adults who
have not had these diseases, particular care should be
taken to avoid exposure.

Reports suggest an apparent association
between use of corticosteroids and left
ventricular free wall rupture after a recent
myocardial infarction; therefore, treatment
with corticosteroids like Dexona must be
used with great caution in these patients.

Contraindications
Dexona is contraindicated in patients with a
hypersensitivity to Dexamethasone Sodium
Phosphate or any other component of this
medication.
This medicine is also contraindicated in cases of
systemic fungal infections unless specific anti-
infective therapy is employed.
Avoid live vaccines in patients receiving immuno
suppressive doses (serum antibody response
diminished).
This medicine should not be administered to patients
with myasthenia gravis, peptic ulcer, osteoporosis or
psychoses

➢Common Generic Dexamethasone Injection
(Dexona) side effects can include Aggression,
agitation, anxiety, blurred vision, decrease in the
amount of urine, dizziness, fast, slow, pounding, or
irregular heartbeat or pulse, headache, irritability,
mental depression, mood changes, nervousness,
noisy, rattling breathing, numbness or tingling in the
arms or legs, pounding in the ears, shortness of
breath, swelling of the fingers, hands, feet, or lower
legs, trouble thinking, speaking, or walking, troubled
breathing at rest and weight gain.

Treatment of Generic Dexamethasone
overdosage is by supportive and
symptomatic therapy. In the case of acute
Dexona overdosage, according to the
patient's condition, supportive therapy may
include gastric lavage or emesis.

Dexamethasone Sodium Phosphate (Dexona 4 mg
injections) has been classified by the US FDA as
Pregnancy Category C.
During animal studies, corticosteroids have been found
to cause malformations of various kinds (cleft palate,
skeletal malformations).
Reduced placental and birth weight have been observed in
animals and humans after long-term treatment. Due to the
possibility of suppression of the adrenal cortex in the
newborn baby after long-term treatment must be
considered, the needs of the mother must be carefully
weighed against the risk to the fetus when prescribing
corticosteroids. However, the short-term use of
antepartum corticosteroids for the prevention of respiratory
distress syndrome, when necessary, does not seem to
pose a risk.
