HIGH PERFORMANCE LIQUID CHROMATOGRAPHY: (HPLC)- It is a technique in analytical chemistry used to sepreate , identify and quantify each content of mixture. It relies on Pumps to pass a pressurized liquid solvent containing the sample mixture trough a column filled with a solid adsorbent material . It was predicted by CALVIN GIDDLINGS,JOSEF HUBER AND OTHERS. PUMPS: The solvent or mobile phase must be passed through a column at high pressure at up to 6000psi or 414 bar. Particle size of S.P. is smaller (5-10 micron) the resistance to flow rate of solvent.
REQUIRMENT FOR PUMPS:- Generation of pressure about 5000psi. Pulse free output & all materials in the pump should be chemically resistant to solvent. Flow rate ranging from 0.1 to 10mil/min. It should be capable of taking the solvent from a single reservoir or more than one reservoir containing different solvents simultaneously. Types: PNEUMATIC PUMP SYRENGE PUMP SINGLE PISTON PUMP DUAL PISTON PUMP
PNEUMATIC PUMP:- In this pump the mobile phase is driven through the column with the use of pressure produced from gas cylinder. It has limited capacity of solvent. Pressure on solvent is proportional to the ratio of piston usually 50:1. About 70ml of the M.P. is pumped from every stroke . ADVANTAGE- Pulse free flow & generates high pressure. DISDVANTAGE: Limited volume capacity Pressure output & flow rate depends on the viscosity & column back pressure.
DISPLACEMENT / SYRENGE PUMP:- Work in the principal of positive solvent pressure. Large stainless steel motor driven hypodermic syringe. Provide constant flow rate Pulse-flow is achieved along with high pressure capability of 200-475 atm. But required a frequent refilling process. Piston driven motor forces the piston down into the reservoir cylinder by means of screw drive. M.P. is forced out through a channel up the center of a screw gear. Two in types: Infusion only pump Infusion withdrawal pump
SINGLE PISTON PUMP: one of the main purpose of hplc pump is to generate a constant flow through the system regardless of the resistance encountered by the M.P. If the pump is consist of a constant head pressure system then the flow would decrease as the resistance in the system increased. hplc pump are mostly piston pump. In the next few screens. We will build a virtual hplc pump and add part as we go along. This single piston pump simply consists of a pump head. Piston and solvent line to a solvent bottle.
DUAL PISTON PUMP: In the previous animation that the solvent flow was erratic. This is because the single piston is either withdrawing the solvent from the bottle or dispensing the solvent of the pump. It is critical in hplc to have constant flow and easiest way to achieve this is to introduce a second piston. THE PURGE VALVE is important part of the pump and is used to direct flow from the autosampler and column while the pump is being primed. Priming the pump involves removing all air in the lines into the pump and removing air in the pump itself.
