Introduction Ibuprofen is a traditional non-steroidal anti-inflammatory drug widely used in the treatment of mild to moderate pain and inflammation. It was discovered by Stewart Adams in 1961. It was used for the treatment of rheumatoid arthritis and osteo arthritis. Formula = C 13 H 18 O 2.
Chemical Structure
Stereoisomerism in Ibuprofen Chiral Center: A chiral center is a carbon atom that is attached to four different groups. Enantiomers of Ibuprofen: (S)-ibuprofen (S-(+)-ibuprofen ). (R)-ibuprofen (R-(−)-ibuprofen ).
R &S Enantiomer
Uses: (Analgesic)Used for pain relief such as headaches, toothaches, back pain, muscle aches, and minor injuries . (Anti-Inflammatory) effective in treating conditions that involve inflammation, such as arthritis. ( Antipyretic ) fever reduction in both adults and children.
Mechanism of Action of Ibuprofen Inhibition of Cyclooxygenase (COX) Enzymes. Ibuprofen primarily exerts its effects by inhibiting the cyclooxygenase enzymes, COX-1 and COX-2 . Reduction of Prostaglandin Synthesis. Analgesic, Anti-inflammatory, and Antipyretic Effects.
Metabolism of Ibuprofen Absorption. Distribution. Hepatic Metabolism . Excretion.
Side Effects of Ibuprofen Gastrointestinal Effects: Nausea, Vomiting, and Diarrhea, Gastric Ulcers. Central Nervous System Effects: Headache and Dizziness. Renal Effects: Kidney Function. Cardiovascular Effects: Increased Blood Pressure
Future Developments in Ibuprofen Development of Safer NSAIDs: Selective COX-2 Inhibitors Improved Gastrointestinal Safety. Combination Therapies: NSAIDs with Gastroprotective Agents IBuprofen with Omega-3 Fatty Acids.
Alternative to Ibuprofen Aspirin. Naproxen. Paracetamol.
