Podophyllotoxin

Podophyllotoxin Podophyllotoxin is the Lactone resin synonym : Indian Podophyllum , Himalayan May apple. Biological source : dried roots and rhizomes of Podophyllum emodi , Podophyllum hexandrum Family : Berberidaceae Other C.C.- podophyllic acid and picropodophyllin , α- peltalin and β- peltalin .

Isolation of podophyllotoxin (method 1) Take the weighed quantity of rhizomes or roots of Podophyllum emodi with methanol. Filter and evaporate to semisolid mass. Dissolve semisolid mass into acidic water. Precipitate is formed which should be allowed for at least for 2hrs

Isolation of podophyllotoxin (method 1) Filter and wash filtrate with cold water. collect the residue, wash with acidified water and dry to obtain dark brown amorphous powder. Extract the residue with hot alcohol. Filter and evaporate to dryness. Re- crystallise the residue in benzene to yield podophyllotoxin

Isolation of podophyllotoxin (method 2) Extract 4gm powder Podophyllum emodi root by stirring and heating with 100ml of ethanol for about 10 minutes. Cool and filter the mixture Re-extract the residue with further 100ml of ethanol Combine the extracts and evaporate to about 20ml

Isolation of podophyllotoxin (method 1) Combine the extract and evaporate to dryness Isolate the Podophyllotoxin by preparative TLC Crystallization at 0C for several days

Identification And Analysis 1. Chemical test – Treat podophyllotoxin with 50% Sulphuric acid it will show violet – blue colour.

Identification And Analysis 2.Thin layer chromatography ( TLC ) Sample preparation – Dissolved about 1mg of podophyllotoxin in 1ml of methanol Standard drug – Podophyllotoxin Stationary phase - Silica gel –G Mobile phase – Chloroform : Methanol (90:10) for about 6 cm (Only glycosides are separated but aglycone like podophyllotoxin remains in the region of the front. 2 nd Mobile phase - Chloroform : Acetone (65:35) Detecting agent - vanillin-sulphuric acid RF value – for 2 nd mobile phase Yellow spot – 0.65

2.Thin layer chromatography ( TLC )

Uses Podophyllotoxin and its derivatives are used as -antiviral agent Antihelminthic Antitumor agents. Podophyllotoxin derived antitumor agents include etoposide and teniposide Used in testicular, breast, pancreatic, lung, stomach, and ovarian cancers.
