Introduction Of Liposome Liposome were first produced in England in 1961 by Dr. Alec D. Bangham . Liposomes are concentric bi-layered vesicle in which an aqueous volume is entirely enclosed by membranous lipid bilayer mainly composed of natural and synthetic phospholipid . The name liposome is derived from two Greek word lipo means fat and soma means body.

Naturally occurring phospholipids Phosphatidylcholine Phosphatidylserine Phosphatidylethanolamine Synthetic phospholipids used are Disteroyl phosphatidylcholine Dioleoyl phosphatidylethanolamine Dioleoyl phosphatidylcholine A- Phospholipids

B- Cholesterol Stabilizes the membrane Act as a fluidity buffer Restrict the transformation of trans to cis conformations

A) Mechanical Dispersion Method A-1 Hand Shaking Method

A-2 Sonication

A-3 French Pressure Cell This Method Is Having The Mechanism Of High Pressure. This Method Will Give Either Uni - Or Oligo -lamellar Liposomes Of Intermediate Size(30-80nm). This Liposomes Are More Stable Compared To The Sonicated Liposomes . But this Method Is Having Some Drawback Of Initial High Cost.

A-4 Micro-Emulsification

A-5 Membrane Extrusion Technique.

This technique is used to process LUVs and MLVs. The size of liposome is reduced by gently passing them through membrane filter of definite pore size at much lower pressure. In this the vesicles content are exchanged with the dispersion medium during breaking and resealing of phospholipids bilayers. As they pass through the polycarbonate membrane they are collected in outlet.

B-3. Reverse Phase Evaporation Method First water in oil emulsion is formed by brief sonication of two phase system containing phospholipids in organic solvent and aquous buffer. The organic solvent are removed under reduced pressure resulting in the formation of a viscous gel. The liposome are formed when residual solvent is removed by continued rotary evaporation under reduce pressure .

Production Cost Is High Short Life Span Leakage & Fusion Of Encapsulated Molecules Low Solubility. Leakage Is Fast. Allergic Reaction May Occurs. Once Administrated Liposomes Can Not Be Removed. Difficult In Large Scale Manufacturing And Sterilization. Disadvantages of liposome

Liposomes are one of the unique drug delivery system, which can be of potential use in controlling and targeting drug delivery. Liposomes are administered orally , parentally and topically as well as used in cosmetics and hair technologies, sustained release formulations, diagnostic purpose and as good carriers in gene delivery. Now a days Liposomes are used as versatile carriers for targeted delivery of drug. Conclusion
