Definition of Apoptosis Apoptosis is defined as a type of cell death that is induced by a tightly regulated suicide program in which cells destined to die activate intrinsic enzymes that degrade the cells’ genomic DNA and nuclear and cytoplasmic proteins.

EXTRINSIC PATHWAY OF APOPTOSIS

It is due to engagement of plasma membrane death receptors . Death receptors belong to TNF aplha family like TNFR1. Death domains are present that deliver apoptotic signals. This pathway can be inhibited by FLIP protein that bind to procaspase 8 and inhivit its activation.

INTRINSIC PATHWAY OF APOPTOSIS

It results from increased permeability of outer mitochondrial membrane with consequent release o f death inducing molecules into cytoplasm. It includes a) Antiapoptotic - BCL2,BCL-X1,MCL1 b) Proapoptotic - BAX and BAK c) Regulated apoptosis inhibitors- BAD,BIM,BID,Puma and Noxa
