Detection of microbes in wastewater Ndou L.F 20006782 NDOU T 21002581 Maluleke M 21007995

INTRODUCTION Wastewater contains a diverse array of microorganisms, including bacteria, viruses, fungi, and parasites. These microorganisms find their way into treated wastewater effluents which poses a risk to the environment and public health. T he detection, identification, and enumeration using microbiological analysis is crucial . Physiochemical analysis of treated wastewater effluents gives insights on their water quality . Our main focus will be based on bacteria and virus such mycobacterium tuberculosis, Escherichia coli, Klebssiella and Human Adenovirus.

OBJECTIVES

SAMPLE COLLECTION AND PREPARATION Sample s will be collected from each WWTP's influent and effluent, upstream and downstream points of the receiving waterbodies. Collected samples will be stored in ice immediately after sampling and then transported to the university of Venda general laboratory pending analysis using microbiological methods. Physicochemical ana lysis of the collected will give insights on the quality of river water. The presence of bacteria wastewater effluent will be established through standard culture methods. Membrane f iltration will be done trap microorganis ms in the 0.22 μm membrane filter paper and c entrifugation will be done to separate substances based on their size, shape and density.
