INTRODUCTION WHAT IS PCV? Packed cell volume (PCV) is the amount of red blood cell, following centrifugation expressed as percentage as the total blood volume PCV can be done in two methods which are; macro and micro hematocrit method. Micro hematocrit: Heparinized capillary is used. In the case of difficulty in drawing sufficient amount of blood this method is used. It is useful particularly in podiatric patients. This is the method I will be taking about. Macro hematocrit: Wintrobe haemotocrit tube is used

PRINCIPLE Blood is centrifuge in a sealed capillary tube and PCV is determine by a special capillary reader. In this case of anticoagulant venous blood obtained from vein puncture, use plain capillary tubes, which are cheaper.

REQUIREMENTS Hematocrit reader Micro hematocrit centrifuge Capillary hematocrit tube Soft wax or sealant Specimen (Blood sample) EDTA bottle

PROCEDURE Method of collecting blood sample : Allow the patients to sit down comfortably Sterilize the site of inoculation and prick with lancet needle or blade Clean the first blood that comes out which may contain blood fluid Using heparinized tube, bend the tube at an angle of 45 fill with blood to ¾ Seal the other side of of the tube with plasticin or sealant Spin at 12000rpm for 5 minutes The results is then read with hematocrit reader in percentage.

RESULTS Normal Range of PCV Adult male – 38%-45% Adult female – 36%-42% Infant – 46%-60%

REASONS FOR CARRYING OUT PCV Incomplete sealing of the tube Double dilution of anticoagulant Method used in reading the result Time and speed of centrifugation FACTORS AFFECTING THE ACCURACY OF PCV To detect anemia as opposed to polyeythemia To test the efficiency of transfusion (post transfusion PCV) To monitor patient placed on hematitinic DISADVANTAGES It cannot be replaced if there is any error

It is necessary to sensitize staff of all various company on how to relate to student by helping them to get the needed knowledge. It would be very important if the organizer of SIWES program could sit with various company and discuss the relevance of the program and to create more IT spaces. Financial aid should be given to student to help them with feeding, transportation etc. All supervisors should visit every student in their various place of attachment RECOMMENDATION

In the course of program, I came to terms with the different behavioural attitude of the working class. The program got a new view on how to handle operational issues with limited time. In all this, I became confident of my ability to think independently. SIWES has improve the practical skills of many students today. Indeed I am glad that I was part of it. The objectives created by the SIWES has been organize to enable student having experience not only the practical aspect of it but also how to co-operate with the member. CONCLUSION
