PYRIDAZINE AND ITS DERIVATIVES Submitted by- Dheeraj Kumar (BP301312)& Harish Santoshi ( BP301315 )

INTRODUCTION Pyridazine is a heterocyclic organic compound with molecular formula C 4 H 4 N 2. It contains six membered ring system with two adjacent nitrogen atoms and is aromatic. It is colourless compound with melting point 208 o c.

PROPERTIES OF PYRIDAZINE CHEMICAL FORMULA :- C 4 H 4 N 2 MOLECULAR MASS :- 80.09 g/mol DENSITY :- 1.11 g/cm 3 BOILING POINT :- 208 c MELTING POINT :- -8 c APPERANCE :- COLOURLESS LIQUID SOLUBILITY IN WATER:- MISCIBLE ISOMERIC FORM:-PYRIMIDINE & PYRAZINE PYRAZINE PYRIMIDINE

SOME DRUGS CONTAING PYRIDAZINE Pyridazine are rare in nature possibly reflecting the scarcity of naturally occurring hydrazine which are building blocks for synthesis of heterocyclic.

DERIVATIVES OF PYRIDAZINE A large number of pyridazine derivatives are synthesised and tested for their pharmacological activity. Some of the important pyridazine derivatives which posses pharmacological activities and studied by us are 3,6-dicholoro-pyridazine and N,N-(diphenyl amino)-pyridazine.

SYNTHESIS OF N,N-(biphenyl amino)-pyridazine :- 1 m mole of 3,6-dichloro pyridazine is mixed with 2 m moles of aniline .The mixture was dissolved in DMF/toluene and refluxed for 8 hours. After 8 hours when the reaction completes, the reaction mixture is recrystallized with suitable solvent.

PHARMACOLOGICAL ACTIVITIES OF PYRIDAZINE DERIVATIVES :- 1) Anti-Inflammatory and Analgesic :- Inhibition of P38 kinase enzyme which is involved in the regulation of TNF and IL . 2)Diuretic Activity:- Some compounds have excellent diuretic activity and different characters from known diuretics like thiazides , carbonic anhydrase inhibitors or furosemide

3) Antiasthmatic Activity :- The pyrido -[2,3-d] pyridazinones were act as potent and selective Type-IV Phosphodiesterase (PDE) Inhibitors These PDE-inhibitors are promising drugs in the management of asthma and inflammation. 4)Antimicrobial Activities:- Infectious diseases caused by microbes like bacteria, fungi and viruses. The purpose of microorganisms is to survive and many of them have determinants of resistance. The anti-microbial effects of some fused pyridazine derivatives showed potential antimicrobial activity.

5)Anticancer Activity :- Cancer in an uncontrolled proliferation of abnormal cells. These mutated cells attack adjoining tissues and sometimes transfer through the blood or lymph distribution to other body organs causing metastases, which are the main cause of death from cancer. A considerable fraction of cancers can be cured by surgery combined with radiotherapy or chemotherapy, particularly if they are detected early. The pyridopyridazine derivatives inhibited cascade of signals which regulate proliferation. Inhibition of proliferation could be achieved by inhibition of cyclin - dependent kinases . Along with above mentioned pharmacological actions pyridazine derivatives also possess antitubecular , antihypertensive and antidiabetic activities.

CONCLUSION Increasing interest in the pyridazines and pyridopyridazines has been observed. These pyridazinne derivatives are showing wide spectrum of biological activities. Recently the pyridazinone moiety has been comprehensively studied in the search for novel and selective drugs. Several biological activities of pyridopyridazines derivatives and may act as important biological active pharmacophores in medicinal chemistry. This moiety may help in exploring new pathways to discover new drugs of potential therapeutic value.
