Leakage Test Fill the container with water Fit with intended closure and keep them inverted at room temp. for 24 hrs. Test is passed if no sign of leakage from container
Collapsibility Test Applicable to containers which are squeezed for removing the contents A container by collapsing inward during use, yield at least 90% of its normal contents at the required rate of flow at ambient temperature
Clarity Aqueous Extract Random selection of unlabeled, unmarked and non laminated portions from suitable containers Cut in strips with total areas 20 sq.cm. Wash the strips for 30 seconds & drain off the water The sample is then taken into flask, previously cleaned with chromic acid and rinsed with distilled water several times Then to flask with sample, 250 ml distilled water is added and autoclave at 121 ͦC for 30 min Same should be done for blank on examination the extract should be colorless & free from turbidity
Water vapour permeability test Fill 5 containers with normal vol. of water and heat seal the bottles with aluminum foil. Weigh accurately each container and allowed to stand for 14 days at relative humidity of 60±5% & temp. 20-25 ͦC. Reweight the container the loss in weight in each container is NMT 0.2%
Transparency test Standard suspension preparation: 1 gm. hydrazine sulphate in 100 ml water and set aside for 6 hr. Take 25 ml. of this solution and add 25 ml. of 10% w/v hexamine solution and stand for 24 hrs. Test solution preparation: Sample is prepared by 16 fold dilution of the standard suspension Fill the containers & cloudiness detectable when compared to a water filled containers. Absorbance is measured at 640 nm and range is within 0.37 to 0.43
Biological Tests Systemic injection test: The test is perform on albino mice. In this 5 mice in each groups i.e test and blank are taken . The sample is inject in mice and observe for any significant biological activities for 4 hrs , 24 hrs , 48 hrs & 72 hrs. if abnormal behaviour such as convulsions & prostrations r loss in body weight ( ˂2 g), the sample does not meet the requirements. 2. Intra cutaneous test: T his test is perform of rabbits. In this reactions like erythema, edema & neurosis are studied for 24, 48, 72 hrs.
3. Eye Irritation test: This test is perform on rabbits. Any irritation in eyes due to sample is studied as compared to blank 4. Resistance to stress: In this the water is acidified with 2M HCL. The acidified water is filled in container and apply force to both end of containers. No break, or deterioration should occur for the sample to pass the test.
