Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR SUPER CRITICAL FLUID EXTRACTION (SFE ) INTRODUCTION: Extraction and isolation of natural products from various sources – generates large amount of waste organic solvents – industrial waste are hazardous to human health- continuous regulation in the use of industrial solvents have increased the amount for super critical fluid technique- SCFE is a clean, safe, inexpensive, non flammable, non toxic, environment friendly solvents. This is the process of separating one components (Extract) from the another components (matrix) using supercritical fluids as the extracting solvent. Super critical fluid is any substance at a temperature and pressure above its critical point, whereas distinct liquid and gas phases do not exists.

Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR Carbon dioxide is the most commonly used supercritical fluids . A fluid is called supercritical if both T and P are above critical values. The state and density of CO2 is shown on pressure- temperature plane. Supercritical fluids are known to provide good reaction media due to their capacity to homogenize reaction mixture, high diffusivity and controlled phase separation and distribution of products. S upercritical fluid (SCF) technology is now considered as A very innovative and promising way to design particles, especially for therapeutic drug formulation. . PRINCIPLE:-

Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR INSTRUMENTATION:- INSTRUMENTATION PART CONSIST OF – Requirement for a supercritical fluid extraction system- Pump (for CO2) E xtraction chamber Recovery Chamber Pressure cell (sample) Means for controlling the pressure Collectin g vessels

Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR PROCESS OF EXTRACTION TECHNIQUE EXTRACTION PROCESS BY SCF- The feed i.e. Powdered solid materials is charged into the extractor . CO2 is fed to the extractor through a high pressure pump (100-350 bar) where the extraction of phytoconstituents takes place. From the extractor CO2, along with the extraction phytoconstituents is sent to the separator (120-50 bar) via pressure reduction valves. At reduced temperature and pressure condition the extract precipitate in the separator . While CO2 free of any extract is recycled to the extractor.

Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR ADVANTAGES:- DISADVANTAGES:- Fast process Improved product properties More economical Environmental friendly Reduce the extraction time Elevated high pressure High power consumption Extraction time is too long Relative high cost of investment

Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR ADVANTAGES FOR THE USE OF CO2 AS A SOLVENT CO2 is inexpensive in nature Non toxic in nature Non flammable Available in high purity Low critical pressure: 73.8 bar Normal boiling point: -78.5°C Temperature : 31.1°C

Dept. of Pharmaceutical Analysis, NIPER-HAJIPUR, Bihar, India NIPER-HAJIPUR APPLICATION OF SUPERCRITICAL FLUID EXTRACTION Extraction and recovery of active ingredients- natural product and in food industries. Extraction of flavors, fragrance and aroma from flowers (jasmine and rose). Extraction of natural food colors( e.g.- curcumin, etc.) Extraction of vitamins, antioxidants, essential fatty acids like B-carotene. Flavor and essence extraction from spices. Removal of pesticides. Removal of alcohol from wine and beer. Decaffeination of coffee
