INTRODUCTION Tetracyclines are a class of antibiotic,which are chemical substances produced by a microorganism that are able to kill other microorganisms without being toxic to the person,animal or plant. Tetracyclines are derived directly from a bacterium known as Streptomyces coelicolor . Tetracycline were discovered in 1940.

MECHANISM OF ACTION Tetracyclines are specific inhibitors of bacterial protien synthesis. They bind to the 30s ribosomal subunit and thereby prevent the binding of aminoacyl tRNA to the mRNA ribosome complex . Tetracyclines also inhibit protein synthesis in the host,but are less likely to reach the concentration required because eukaryotic cells do not have a tetracycline uptake mechanism.

SPECTRUM OF ACTIVITY Tetracyclines are broad spectrum antibiotics,active against Gram positive and Gram negative bacteria,spirochetes,mycoplasma,rickettsie and chalmydiae .

Mechanism of resistance Cell become resistance to tetracycline 3 mechanisms Enzymatic rarest type of resistance,where an acetyl group is added to the molecule,causing inactivation of drug. Efflux resistance gene encodes a membrane protien that actively pumps tetracycline out of cell. Ribosomal protection which blocking tetracycline from binding to the ribosome.

ADMINISTRATION Oral capsules is the dosage form in which tetracyclines are most commonly administered. The capsule should be taken 1/2hr before or 2hr after food. Not recommended for i.m . route of administration .
