Theory of this practical : Ferrous phosphate syrup is prepared by chemical reaction between iron and phosphoric acid and other electrolyte supplements like calcium carbonate , potassium bicarbonate and sodium phosphate. The electrolytes such as calcium (Ca++) , Sodium (Na++) and Potassium (K+) are also supplied in phosphate form. In this preparation, half of portion of phosphoric acid reacts with iron and forms ferrous acid phosphate. Remaining half of the portion of phosphoric acid reacts with calcium carbonate, potassium bi carbonate and sodium phosphate and forms corresponding phosphates. But the amount of phosphoric acid is insufficient to complete this reaction. Hence the excess of acid phosphate present in ferrous acid phosphate {Fe(H 2 PO 4)2 } is used to complete this reaction.
The following reactions takes place, during the preparation of ferrous phosphate syrup. Fe + H 3 PO 4 = Fe(H 2 PO 4 ) 2 + H 2 O Iron Phosphoric Acid Ferrous acid phosphate CaCO 3 + 2H 3 PO 4 = Ca(H 2 PO 4 ) 2 + CO 2 + H 2 O Calcium Carbonate Calcium dihydrogen phosphate KHCO 3 + 2H 3 PO 4 = KH 2 PO 4 + CO 2 + H 2 O Potassium Carbonate Potassium dihydrogen phosphate Na 2 HPO 4 + 2H 3 PO 4 = NaH 2 PO 4 + CO 2 + H 2 O Sodium Phosphate Sodium dihydrogen phosphate Cochineal is used as colouring agent, which is obtained by aqueous extraction of cochineal . In this preparation it is used as masking agent to mask, if any colour change of the preparation occurs , due to oxidation of constituents during storage. Orange flower water is used as flavouring agent. Sucrose is used to prepare syrup, which is used as sweetening agent to mask the specific iron taste of the preparation. It also helps in giving viscous nature to the preparation and also acts as preservative, because of its higher concentration.
Requirements : Apparatus : Weighing Balance, Beaker , Stirrer , conical flask, measuring cylinder , water bath etc. Chemicals : Iron phosphoric acid, Calcium Carbonate , Potassium Coarbonate , Sodium Phosphate , Cochineal , Sucrose , Orange flavour water of commerce, Undiluted and Purified water.
Procedure : Clean all the glassware and dry them properly as per sop. Dilute half the quantity of phosphoric acid with little amount of water and iron to it. Gently heat this solution on a water bathn until iron gets dissolved. In a separate beaker transfer remaining portion of phosphoric acid and add calcium carbonate, potassium bi carbonate, sodium phosphate with continuous stirring. All step CO 2 will release and allow CO 2 to evolve completely.
Mix both the solution with stirring and allow to stand for few minutes to complete the reaction in a conical flask. Boil cochineal in water for 15 min. Cool , strain and pass sufficient water through the stainless. To this add sucrose and again boil for 15 minutes to form colored syrup. Filtered iron slution into the colored syrup , to add orange flavoured water to pass sufficient water through the filter to produce the required volume to mix allow standing for cooling and filter of necessary. Transfer preparation for narrow mouth coloured glass bottle clot , to close with cap label and submit.
Category : Iron and Electrolyte (Calcium, potassium, sodium) supplement in anemic patient. Storage : Store in well closed container in cool and dark place. Result : 10 ml of compound syrup of Ferrous Phosphate BPC 1968 preparation is submtted in well closed amber colored container with neat label.
