WET MOUNT METHOD

In a wet mount, the specimen is placed in a
drop of water or other liquid held between the
slide and the cover slip by surface tension.
This method is commonly used, for example,
to view microscopic organisms that grow in
pond water or other liquid media, especially
when studying their movement and behavior.



Advantages of a wet mount

No artefact: direct microscopy
makes living or-ganisms and
biological structuresclearly and
immediately visible,without
artifacts due to dehydration,
fixation and staining used for
processing permanent mounts
the may alter the morphological
characteristics and modify the
physicochemical properties of
the matter.
Quick preparation. Observation
of movements of protozoas
and cell organelles such as cilia
in ciliated endocervical cells.

Disadvantages of a wet mount

• Storage: wet mounts cannot be
stored over a long time because the
specimen dries up. This problem
may be easily solved by taking
photos of the microscopic images
using a digitalcamera system
applied to the phase contrast
microscope.
• Difficulty in taking pictures due to
the movement of protozoas or
ciliated endocervical cells. However
it is possible to film the moving
organisms using the digital camera
system or wait a few minutes for the
movements to fade or disappear.




Types of Wet mount method

1.Normal Wet drop method

Use a dropper to place One drop of water in the
centre of slide.
Pace a small thin section of the sample on the top
of the drop.
Place a cover slip over the sample angling it like a
hing.

Normal Wet drop method

Use normal slide.
Small sample size is OK.
Only good for short term
use.
Only show Brownian
motion.




Hanging drop method

Use depression slides.
Larger sample size
needed.
OK for longer term use.
Show the both
Brownianas well as true
motion.
