Introduction Alzheimer’s disease is a progressive neurodegenerative disorder characterized by memory loss, cognitive decline, and personality changes.

Epidemiology • Over 50 million cases globally • Prevalence increases with age • More common in women • Expected to triple by 2050 due to aging population

Etiology (Causes) • Genetic: APP, PSEN1, PSEN2 mutations • Environmental: head trauma, toxins • Lifestyle: inactivity, poor diet, smoking • Age: strongest risk factor

Pathophysiology • β-amyloid plaques form outside neurons • Tau protein tangles form inside neurons • Leads to neuron death and brain shrinkage, especially in the hippocampus

Symptoms • Memory loss, disorientation • Language and judgment impairment • Personality and mood changes • In late stages – loss of mobility, dependency

Diagnosis • Cognitive tests (MMSE) • Imaging: MRI, PET scans • Biomarkers: β-amyloid, tau in CSF • Rule out reversible causes of dementia

Treatment • Cholinesterase inhibitors: Donepezil, Rivastigmine • NMDA antagonist: Memantine • Supportive care: exercise, diet, cognitive therapy

Recent Research • Monoclonal antibodies (Aducanumab) • Stem cell and gene therapy research • Lifestyle interventions for prevention • Early detection biomarkers in focus
