Tetraisopalmitate ascorbate(abbreviation VC-IP),It is an esterified
vitamin C derivative and a new generation of derivatives.
Other names: tetrahexyl decanol ascorbic acid ester, esterified VC, fat
soluble vitamin C.
Take the letters in its English name:Vitamin C tetra-isopalmitate,
abbreviation VC-IP;
INCI :Ascorbyl Tetraisopalmitate

Prototype VC has several disadvantages:
•Poor stability, easy to discolor and oxidize under air and light
•VC must be kept below pH 3.5 and is therefore irritating to the
skin
•Because VC is water-soluble, but our skin has a "strong" lipid
bilayer, so this makes it difficult to work through the skin.

STABILITY OF VC-IP
VC-IP Store at 40 ℃ or 60 ℃ for 1 month. There is no obvious
change in appearance and hue. The measured vc-ip is heated
at the rate of 5 ℃ / min at 200 ℃, and its weight remains
unchanged at 30-500 ℃. These results show that vc-ip has
good thermal stability (Fig. 1)

homogenized and extracted under
the conditions of 3%
metaphosphoric acid and 0.2%
ethanol, and then separated by
high performance liquid
chromatography on ODS column。

in cells
• about 14 times than VC itself. After
lipolysis, vc-ip is transformed into VC in
skin cells, so as to play the role of VC。

test method:
Human melanoma cells (hm-3-ko) were cultured
with VC-IP in the concentration range of 0% - 0.2%
for 4 days. The cells were then collected with
trypsin and centrifugation. After the cells were
counted, the melanin density of the cells was
measured by Marcia laser densitometer.
VC-IP inhibits tyrosinase activity and
melanin production
Compared with the control group, the
melanin synthesis of VC-IP decreased
from 72% to 12%

Use the serum-free conditioned medium was cultured in
50um with or without VC-IP for 48 hours, the NHDF cells
were ultrafiltration and concentrated on a SDS-
polyacrylamide gel containing 0.2% gelatin under non
reducing conditions, followed by Coomasie Brilliant Blue
R250 staining, and then measured with a laser densitometer。
VC-IP significantly inhibited the activity of MMPs and the
increase of collagenase induced by UV irradiation
When VC-IP (20-50um) was used, the
activities of MMP-2 and MMP-9 degrading
gelatin were inhibited to 18% - 23% and
10% - 17% respectively. The activity of
MMPs in VC (20-50um) group was 39% -
46% and 42% - 52% respectively.

test method :
Human skin fibroblast NHDF cells
were given L - [2,3h] proline for 24
hours in the presence or absence of
vc-ip or VC (20-50um), followed by
cell lysis and subsequent
neutralization by NaOH treatment.
The obtained cell extract was
thoroughly hydrolyzed with
collagenase of Clostridium
perfringens, centrifuged after
tcatennin treatment to produce
supernatant, precipitated into
collagen and non collagen
components, and evaluated by
Aloka liquid scintillation counter lc-
3600 SCI.
The decrease of collagen is manifested
in the photoaging of human skin. It is
reported that the degradation of
collagenase is accelerated due to the
release of collagenase from infiltrating
cells induced by ultraviolet light.
In this case, the degradation rate of
collagen exceeds the rate of
biosynthesis, resulting in the reduction
of collagen.
The effect of VC-IP on promoting collagen synthesis
is about 3.5 times than VC.
VC-IP increased collagen synthesis of
human skin fibroblasts by 170% - 195% in
the concentration range of 20-50um, while
VC increased collagen synthesis by 112% -
127% in the same concentration range。

Reference formula

•VC-IP is soluble in most oils (whether
polar or nonpolar), except Polyols (such
as glycerol, propylene glycol,
butanediol). VC-IP can be quickly
dissolved in ethanol, hydrocarbons,
esters and vegetable oils, such as
propylene glycol monooctanoate,
castor oil, glycerol triisooctanoate
(glycerol triethylhexanoate), olive oil,
cetyl alcohol ethyl hexanoate and
mineral oil。
•The pH value of the designed formula
(in aqueous phase) shall not exceed 6.
Although VC-IP has good oil solubility,
its stability will be affected by pH value.
Therefore, it is recommended to adjust
the pH value below 6.0 before adding
VC-IP (the pH value cannot be adjusted
after adding). Vc-ip can also be used in
formulations with pH 7 when used in
combination with chelating agents and
antioxidants。
