Lesson Objective: Identify the parts of a compound microscope and the function of each part.

What is a Compound Microscope? It is a type of microscope that uses visible light to illumine the object, being observed. It consists of two optical parts, the ocular lens in the eyepiece and the. objective lenses

The parts of a Compound light microscope can be classified into: Mechanical Parts hold the different parts together and are used to adjust the magnifying and illuminating parts to focus on a specimen. Magnifying Parts are involved in enlarging the image of a specimen. Illuminating Parts parts are used to collect light and ate a clear image.

MECHANICAL PARTS PARTS DESCRIPTIONS AND FUNCTION Draw tube a small cylinder attached to the upper part of the body tube that holds the ocular lens Body tube connects the ocular lens to the revolving nosepiece Coarse adjustment knob An adjustable knob that moves either the body tube or stage upward or downward in greater increments to bring the specimen into initial focus; should be used only with the scanner or low power objective Fine adjustment knob An adjustable knob that moves either the body tube or stage upward or downward in lesser increments to bring the specimen into sharp focus; preferably used when focusing using the high power or oil immersion objective Arm supports the body tube and is used to carry the microscope Revolving nosepiece a circular and revolving part connected to the body tube that holds the objectives Stage a platform where the specimen slide is placed; has a hole at the center for the light to pass through the specimen Stage clips used to secure the specimen slide on the stage Inclination joint attaches the arm to the pillar; used for tilting the microscope Pillar provides support above the base and where the arm is attached Base provides firm and steady support to the entire microscope

MAGNIFYING PARTS PARTS DESCRIPTIONS AND FUNCTION Ocular lens or eyepiece a detachable cylinder located on top of the draw tube that is used to view the specimen; usually capable of magnifying objects up to 10x; may have a black line that serves as a pointer Objective lens used to magnify the specimen under study A compound microscope usually has three objective lenses: low power objective (LPO), high power objective (HPO), and oil immersion objective (010). Some microscopes have an additional scanner objective. In some microscopes, the Scanner replaces the Ol0. Objective lenses are always used in the following order: scanner (if present), LPO, HPO, and OI0.

ILLUMINATING PARTS PARTS DESCRIPTIONS AND FUNCTION Condenser concentrates light onto the specimen; located above the iris diaphragm; may have knobs to adjust the condenser’s position Iris diaphragm regulates how much light passes through the specimen; can be controlled using a lever that can be moved back and forth Mirror reflects light and directs it to the specimen slide, then reflects the image back to the eyes; found below the stage near the base
