INTRODUCTION Used to identify Anthrax bacillus in microscope. Actually a Staining technique. Employed for the presumptive diagnosis of anthrax in animals. Stains the polypeptide capsular material of anthrax bacilli that is composed of poly-d–Glutamic Acid.
PROCEDURE Take blood from tip of ear and make a impression slide. This is stained with Poly Chrome Methylene Blue (PCMB ) for 30 seconds. Examine under microscope. If the sample is positive, an amorphous purplish material is noticed around the blue bacilli. This purplish material is capsule around blue bacilli body.
