Long chain polysaccharides obtained from seaweed algae Is mixture of two polysaccharides such as aggarose (70%) and agaropectin (30%) Properties of agar: Acts as good solidifying agent It has no nutritional value in media Stable at different temperatures used for microorganisms Melt at 95 to 98 degree Celsius and remains liquid upto 40 to 42 degree Celsius Easily available and economical

Water Tap, pure distilled water may be used for preparation of culture media by dissolving various organic and inorganic compounds All enzymatically controlled chemical reaction occurs within cell in presence of water Copper distilled water cannot be used for preparation of culture media because copper inhibits bacterial growth Essential for growth of all microorganisms

Indicator media These media contain an indicator which changes colour when a bacterial species grows in them E.g. Wilson and Blair medium Salmonella typhi reduces sulphite to sulphide in presence of glucose and the colonies have black metallic shine

Enrichment media When specific substance is added in a liquid medium which inhibits the growth of unwanted bacteria and favors the growth of wanted bacteria such media called enrichment media e.g. Tetrathionate broth, selenite F broth

Sugar media In sugar medium, 1% sugar is added to peptone water with a suitable indicator Durhams tube (small tube) is kept inverted in the tube containing this medium to detect gas production

Assay media: These media have specific composition and are used for the assay of antibiotics, amino acids and vitamins Storage media: This media help in. Preservation and storage of bacteria for long period e.g. Nutrient agar slots, blood agar slants etc.
