Definition:
Qualitybydesign(QbD)isasystematicapproachtoproductdevelopmentthat
beginswithpredefinedobjectivesandemphasizesproductandprocess
understandingandcontrolsbasedonsoundscienceandqualityriskmanagement
(ICHQ8).
QbDisaconceptfirstdevelopedbythequalitypioneerDr.JosephM.Juran.He
belivedthatqualityshouldbedesignedintoaproduct,andthatmostqualitycrises

3.Critical Quality Attributes (CQAs):
Itisdefinedas“aphysical,chemical,biologicalormicrobiologicalpropertyor
characteristicsthatshouldbewithinanappropriatelimit,range,ordistributionto
ensurethedesiredproductquality.
CQAsaregenerallyassociatedwiththedrugsubstance,excipients,intermediates
anddrugproduct.
Thequalityattributesofadrugproductmayincludeidentity,assay,content
uniformity,degradationproducts,residualsolvents,drugrelease,moisture
content,microbiallimits.
Physicalattributessuchascolor,shape,size,odor,scoreconfiguration,and
friability.
Theseattributescanbecriticalornotcritical.

4.Critical Material Attributes (CMA):
CMAofadrugsubstance,excipient,andin-processmaterialsisaphysical,
chemical,biological,ormicrobiologicalcharacteristicofanrawmaterialthat
shouldbeconsistentlywithinanappropriatelimittoensurethedesiredqualityof
thatdrugsubstance,excipient,orin-processmaterial.
TheCMAislikelytohaveanimpactonCQAofthedrugproduct.
Amaterialattributescanbeanexcipientsrawmaterial,drugsubstances,reagents,
solvents,packaging&labelingmaterials.

5.Critical Process Parameters (CPP):
ACPPofmanufacturingprocessaretheparameterswhich,whenchanged,can
potentiallyimpactproductCQAandmayresultinfailuretomeetthelimitofthe
CQA.
Thisisdependonoperationalparameterofasystemorunitoperation.(Mixing
speed,flowrate,mobilephaseratio,molarityofbuffer,pHofbuffer)andprocess
statevariables(Temp,Pressure,viscosity,density,angleofrepose)
