BACKGROUND
Stoma(plural-stomata)isaminuteepidermalopeningcoveredbytwokidneyshaped
guardcellsindicotleaves.Theseguardcells,inturn,aresurroundedbyepidermal
(subsidiary)cells.Stomataperformthefunctionsofgaseousexchangeandtranspirationin
plants.Thenatureofthestomata,aswellas,thestomatalindexandstomatalnumberare
importantdiagnosticcharacteristicsofdicotleaves.Stomatalnumberisdefinedasthe
averagenumberofstomatapersqmmofepidermisoftheleaf.Theactualnumberof
stomatapersqmmmayvaryfortheleavesofthesameplantgrownindifferentenvironment
orunderdifferentclimaticconditions.Itis,howevershownthattheratioofthenumberof
stomatatothetotalnumberofepidermalcellsinagivenareaofepidermisisfairlyconstant
foranyageoftheplantandunderdifferentclimaticconditions.

PROCEDURE
PreparationoflaminaTakeamatureleaf.Iftheleafissmall,thewholeleafmaybetakenandifthe
leafislarge,cut5mmsquarepiecesfromthemiddleportionbetweenthelaminaandmidrib.
Freshleaf
Sometimestheepidermiscanbeeasilypeeledoffinthickleavesbybreakingitintopiecesby
sheeringaction.Separatetheepidermisandtreatwithchloralhydrate.
Cutanumberof5mmpiecesfromthemiddleportionbetweenthelaminaandmidrib.
Boilwithchloralhydrateinatesttubeplacedinawaterbath.Theepidermisseparatesout.
Carefullyplacethe
epidermisonaslidewiththehelpofabrushalongwith1-2dropsofchloralhydrate;coolandthen
placeacoverglass.

PROCEDURE
Prepareanimprintoftheepidermis:
Takealittlepieceofgelatingel(50%)withthehelpofaneedle.Smearitonahotslide,
placeafreshleafandslightlypresstheleaf.Inverttheslideandcoolitunderawatertap
tillthegelissolidified.
Thentheleafisremoved.Thisleavesanimprintofthestomataandepidermalcellsonthe
gel.
Tracetheepidermalcellsandstomatawiththehelpofcameralucida.
