FERMENTATION OF STREPTOMYCIN Test Organism - Streptomyces griseus

REQUIREMENTS Substrate-glucose, fructose, galactose , xylose , mannitol , lactose , starch Source of nitrogen- peptone,meat extract, soy extract, the residues from alcohol distillation, dried pencillium mycelium, ammonium salts,nitrates , glycine

Growth stimulating agents-phenyl acetic acid, l – naphthalene Antioxidant-starch , agar , sodium sulphite

3. PHASES first phase[2 days] consist of mycelial growth requirements oxygen, glucose, nitrogen, phosphorous pH rises to 8.0 due to production of ammonia

Second phase(1 to 2 days) In this streptomycin is produced without the production of mycelium Large quantities of glucose and oxygen are required Ammonia is utilized and ph falls to about 7.0
