The Pentose Phosphate Pathway Dr. Oligo Asoka Jacob Senior Pharmacist B.Pharm MHSCM Dr. Oligo Asoka Jacob, B.pharm, MHSCM

Introduction Also known as the Hexose Monophosphate (HMP) Shunt. It is an alternative route to the metabolism of glucose. The primary function of this pathway is to: Form NADPH for the synthesis of fatty acids and steroids. Form ribose-5-phosphate for the synthesis of nucleotides and nucleic acids. The primary role of the PPP is anabolic because it involves the formation of NADPH used in anabolic reactions and ribose-5-phosphate used in nucleic acid synthesis. Dr. Oligo Asoka Jacob, B.pharm, MHSCM

Introduction The pathway takes place in the cytoplasm of cells in the liver, adrenal cortex, and lactating mammary glands. The pathway has two phases: Irreversible Oxidative phase: NADPH is generated Non-reversible oxidative phase: synthesis of 5-carbon sugars. Dr. Oligo Asoka Jacob, B.pharm, MHSCM

PPP Dr. Oligo Asoka Jacob, B.pharm, MHSCM

Oxidative phase Dehydrogenation of glucose-6-phosphate to 6-phosphogluconolactone catalyzed by glucose-6-phosphate dehydrogenase. Hydrolysis of 6-phosphogluconolactone to 6-phosphogluconate by the enzyme lactonase. Decarboxylation follows with the conversion of 6-phosphogluconate into ribulose-5-phosphate. Dr. Oligo Asoka Jacob, B.pharm, MHSCM

Non-oxidative phase Dr. Oligo Asoka Jacob, B.pharm, MHSCM
