Introduction The urine culture is used to diagnose a  urinary tract infection (UTI) and to identify the pathogenic  causing the infection. Germs can infect wounds and cause pain, swelling, warm, and redness around the wound and can lead to the formation of pus .

Objective To test and analyze urine sample, pus sample , stool culture and blood culture to identify the microorganisms causing infection (if present) and performing antibiotic sensitivity test (AST).

METHODOLOGY COLLECTION OF THE SAMPLE FOR URINE / STOOL CULTURE: 20-30ml of the sample is collected in a sterile plastic container. FOR PUS SAMPLE : A sterile cotton swab is used to collect pus  from a wound site.

Plating and streaking A fter the collection of the sample it is streaked on first the macConky agar plate then onto the nutrient plate. Incubation

ANTIBIOTIC SUSCEPTIBILITY /SENSITIVITY Colony of a plate is mixed thoroughly with sterile saline solution . Streaked with cotton swab on a MHA PLATE or nutrient agar plate . Antibiotic discs are placed gently with the help of forceps.

Result

Conclusion ANTIBIOTIC SENITIVITY is done only when the results are positive that is there is presence of infection . IDENTIFICATION is a very important step Like shiny colonies , rough colonies and colorful colonies etc. Antimicrobial susceptibility tests (ASTs) basically measures the ability of an antibiotic or other antimicrobial agent to inhibit the invitro microbial growth.
