Calcitonin
•Peptide hormone
•Parafollicular cells or clear (C) cells of thyroid gland synthesize and
secrete calcitonin
•Calcitonin is stored in the secretory vesicles

Stimulus of secretion of calcitonin
•High serum Ca
2+
levels
High serum Ca2+ level
Calcitonin release
Acts on bones and kidney
Decrease serum Ca2+
level

Physiological actions
•On bone-osteoclast possess receptors for calcitonin
•Inhibits bone resorption
•On kidneys-decrease Ca
2+
and phosphate reabsorption
•Calcitonin levels are increased during pregnancy in order to prevent the
bone loss in mother (bone formation and lactation are major Ca2+
draining events)

Mechanism of action of calcitonin
•Calcitonin binds to GPCR which may activate either adenylyl cyclase or
phospholipase C
•Activation of protein kinase C and increase intracellular Ca
2+
concentration
•Release of calcitonin containing vesicles via exocytosis.
