*CAP test:
-Cellulose acetate precipitate>>>>>Serology test : -Ag + Ab =
reaction( precipitation).
*Aim:to detect the E.histolytica antigen in patientserum.
*Requirements:
1/plastic box contain spongy.
2 /Template.
3/Cellulose Acetate paper
4/soluble amoebic antigen
5/Phosphatebuffer saline.
6/Negative control.
7/Posative control.
8/Nigrosin stain.
*Procedure:
1/Immerse the paper ( CAP ) in Phosphate buffer saline.
2/By template make 5 well in paper.
3/With fine capillary place one drop ( 5ul ) of Pt serum and
positive , negative control also placed . 3ul of amoebic antigen
placed in central well.
4/Take the paper at room temperature for 4 hour.
5/Wash the paper in saline 5(minute).
6/Immerse the paper ( CAP ) in Nigrosin stain for 2 mint and
Wash the paper in water.
7/Look for precipitin arch Entamoeba histolytica.
Advantages of CAP test:
1/ It is very durable.
2/ Resistant to temperature changes.
3/ Comfortable to wear for a long period of time.
4/ Hypoallergenic and renewable.
5/ Can be painted easily.
Disadvantages of CAP test:
1/High moisture absorption.
2/Dimensional instability.
