Acetate Injection) is s a progestin injectable contraceptive
which is used to prevent pregnancy in women 18 years and
above.
This medication works by preventing the growth and release
of an egg (ovulation) during the menstrual cycle.
Medroxyprogesterone acetate hinders (in the usual dose
range) the secretion of pituitary gonadotropin which, in turn,
prevents follicular maturation and ovulation.

Uses of Depo-Provera Contraceptive Injections
Depo Provera Contraceptive Injection is a well-known brand name for
Medroxyprogesterone Acetate, a contraceptive injection that contains the

Recommended Dosage
The dosage of Depo-Provera Contraceptive Injections
(Medroxyprogesterone Acetate Injection) is given below:
The recommended dosage of Depo-Provera Contraceptive Injection
is 150 mg of Depo-Provera CI every 3 months (13 weeks)
administered by deep IM injection in the gluteal or deltoid muscle.
The 1 mL vial as well as the 1 mL prefilled syringe of Depo-
Provera CI should be vigorously shaken just before use to ensure
that the dose being administered represents a uniform suspension.

Contraindications
Depo-Provera Contraceptive Injection is contraindicated in persons
with a hypersensitivity (allergy) to Medroxyprogesterone Acetate
or any of the other ingredients of this medicine.
Depo-Provera CI is also contraindicated in known or suspected
pregnancy or as a diagnostic test for pregnancy, Active
thrombophlebitis, or current or past history of thromboembolic
disorders, or cerebral vascular disease, Known or suspected
malignancy of breast, significant liver disease and undiagnosed vaginal
bleeding.

Before Taking This Medicine
You may need to have a negative pregnancy test before you start Depo-
Provera.
You should not use Depo-Provera if you are pregnant, or if you have:
abnormal vaginal bleeding that has not been checked by a doctor, liver
disease, breast cancer; or a history of stroke or blood clot.
Make sure Depo-Provera is safe for you, tell your doctor if you have ever had:
light or irregular menstrual periods; risk factors for osteoporosis (such as low
bone mineral density, a family history of osteoporosis, drinking large amounts
of alcohol, or if you smoke); a breast lump, an abnormal mammogram, or
bleeding from your nipples; kidney disease; high blood pressure; breast
cancer (in you or a family member);

Before Taking This Medicine
You should not use Depo-Provera if you are pregnant, or if you have:
diabetes; depression, or an eating disorder; seizures; asthma or
migraine headaches.
Tell your doctor right away if you become pregnant, or if you plan to
become pregnant soon after you stop using Depo-Provera CI.
It may not be safe to breastfeed while using Depo-Provera. Ask your
doctor about any risk.

Storage Instructions
Depo-Provera Contraceptive Injection
(Medroxyprogesterone Acetate Injection) have to
be stored at controlled room temperature i.e. from
20°C to 25°C (68°F to 77°F) with excursions
permitted between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away
from children and pets.

Warnings
Loss of Bone Mineral Density: Use of Depo-Provera CI decreases serum estrogen
levels and is associated with significant loss of bone mineral density (BMD) as
bone metabolism accommodates to a lower estrogen level.
Bleeding Irregularities: Most women using Depo-Provera Contraceptive Injection
experience disruption of menstrual bleeding patterns. Changed menstrual
bleeding patterns also include irregular or unpredictable bleeding or spotting, or
rarely, heavy or continuous bleeding.
Thromboembolic Disorders: The doctor must be alert to the earliest
manifestations of thrombotic disorders (thrombophlebitis, pulmonary embolism,
cerebrovascular disorders, and retinal thrombosis).
Ocular Disorders: Depo-Provera must not be readministered pending
examination in case there is a sudden partial or complete loss of vision or if there
is a sudden onset of proptosis, diplopia, or migraine.

Warnings
Unexpected Pregnancies: To make sure that Depo-Provera CI is not
administered inadvertently to a pregnant woman, the first injection must be given
ONLY during the first 5 days of a normal menstrual period; ONLY within the first
5-days postpartum if not breast-feeding, and in case exclusively breast-feeding,
ONLY at the sixth postpartum week.
Ectopic Pregnancy: Doctors must be alert to the possibility of an ectopic
pregnancy among women using Depo-Provera CI who become pregnant or
complain of severe abdominal pain.
Lactation: Detectable amounts of Medroxyprogesterone Acetate have been
identified in the milk of mothers receiving Depo Provera Contraceptive Injection.
Anaphylaxis and Anaphylactoid Reaction: Anaphylaxis and anaphylactoid
reaction have been reported with the use of Depo Provera Contraceptive Injection.
In cae an anaphylactic reaction occurs appropriate treatment should be started.
Serious anaphylactic reactions require emergency medical treatment.

Overdose
In case of patients with a mild overdose, it is possible that no symptoms
may occur. Depo-Provera CI overdose Symptoms are more likely to occur
with high doses, or when too much Depo-Provera is used over a long
period of time.
Symptoms of an overdose can includeHeadache, Dizziness, Yellowing
of the skin or the whites of the eyes (jaundice), Irregular menstruation,
such as spotting or heavier or lighter periods, Breast pain, Bloating,
Depression and Decreased libido (sex drive). Treatment for a Depo-
Provera overdose will normally involve supportive care, which consists of
treating the symptoms that occur as a result of an overdose.
