Basic Components Of TDDS Drug substance Polymer matrix Penetration Enhancers Adhesives Backing Laminates Release Linear Other Excipients

Drug substance For successfully developing a transdermal drug delivery system, the drug should have following property: Low molecular weight Affinity for both lipophilic and hydrophilic phase Low melting point Short half life and non irritating

Polymer Matrix Permeation Enhancer Role: Control the release rate of drug Examples of polymers: Natural: gelatin, cellulose derivatives, gums etc. Synthetic Elastomer: neoprene, silicone rubber etc. Synthetic Polymer : polyethylene, polyvinyl alcohol, polypropylene etc. Role: Substance that increase skin permeability, allowing the drug to pass through the stratum corneum. Examples: ethanol, dimethyl sulfoxide , oleic acid, glycols, essential oils, trepenes etc.

Adhesives Baking Laminates Purpose: Ensures the patch to adheres to the skin. Types of Adhesives: Acrylic, silicone-based, and polyisobutylene adhesives. Properties: non-irritating, maintain adherence over the application period , and allow controlled drug release. Role: Provides support and protection for the drug formulation and prevent external moisture or contaminants from affecting the patch. Materials: Polypropylene, polyester films.

Release Liner Role: protects the patch before use and is removed before application. Materials: silicone-coated paper or polyethylene-coated paper

Additional Excipients Plasticizers: Improve flexibility of the polymer matrix. Examples: dibutyl phthalate, triethyl citrate. Stabilizers: prevent degradation of the drug. Examples: antioxidants, UV absorbers.
