Ph meter Method: Operation of pH meter : Prepare the sample : First, prepare the sample that you want to test. This may involve diluting the sample with distilled water or making some other preparation. Calibrate the pH meter : Before you begin testing, you should calibrate the pH meter to ensure that it is giving accurate readings. To do this, you will need to use a calibration solution of known pH. Follow the manufacturer’s instructions for calibrating the pH meter. Insert the electrodes : Once the pH meter is calibrated, insert the electrodes into the sample chamber or the sample itself. Make sure that the electrodes are fully immersed in the solution.
Ph meter Wait for the reading to stabilize : It may take a few minutes for the pH meter to stabilize and give a reading. Wait for the reading to stabilize before recording the pH value. Record read ings : Once the reading has stabilized, record the pH value. If the pH meter has a digital display, you can simply read the value off the screen. If it has an analog meter, you will need to read the value from the scale on the meter.
Calibration of pH Meter Preparation of buffer (ph7.0 ) Add 100 ml 0.1 M KH2PO4 in a beaker containing about 50 ml of distilled water, then slowly add 58.2 ml of 0.1 M NaOH and make up the volume up to 200 ml with distilled water, stir well for proper mixing. Preparation of buffer (ph4.2) • Add 100 ml 0.1 M potassium hydrogen phthalate in a beaker containing about 50 ml of distilled water, then slowly add 6.0 ml of 0.1 M NaOH and make up the volume up to 200 ml with distilled water, stir well for proper mixing. Preparation of buffer (ph9.6) 100 ml 0.025 M Na2B4O7.10H2O (borax) in a beaker containing about 50 ml of distilled water, then slowly add 22.2 ml of 0.1 M NaOH and make up the volume up to 200 ml with distilled water, stir well for proper mixing.
Calibration of pH Meter… Method : 1 . Switch on the pH instrument, wait for at least 5 minutes (electrode dipped in distilledwater ). 2. Take out the electrode from distilled water, dry with lint free tissue paper by gentle rubbing ensuring no particle remains in electrode. 3. Dip the electrode in buffer with pH 7.0, wait for few minute (for stabilization) and press the calibrate button key of the instrument. After getting stabilized reading press measure key so that instrument will set value of the buffers pH.
Calibration of pH Meter.. 4.Remove the electrode, rinse with distilled water, and repeat the above procedure with other two buffers. The instrument is calibrated and is ready for measuring pH of samples. After taking pH readings of samples clean and dip the electrode with distilled water
Calibration of pH Meter… Learning outcomes: A pH meter is an essential tool in laboratory settings to measure the pH of solutions in pharmaceutical, chemical, and biotechnology industries. A laboratory pH meter has a high measurement range, providing the user with a more accurate and precise reading. A pH meter is also used to monitor the growth of pathogenic microorganisms that may spoil food or beverages, or negatively impact the taste and quality of the product.
