INTRODUCTION INCLUSION BODIES IN RED BLOOD CELLS Inclusion bodies are those things that just appear in cells and do not really do anything .They are nuclear /cytoplasmic aggregates of stainable substances usually proteins. Inclusion bodies in red blood cells are almost indicative of some sorts of pathology & thus , it is useful to understand each inclusion bodies. They are Howell Jolly bodies , Heinz bodies ,Hemoglobin H Inclusion, pappeinhemier body, Basophilic stippling and Cabot rings

HOWEL –JOLLY BODIES These are histopathological findings on a blood smear .They appear as a very dark purple spot within cytoplasm of red blood cells. They are basophilic nuclear remnants or remnants of DNA. They are round to oval shaped,1um across ­& usually restricted to only 1 per cell. If Howell-Jolly bodies are present in high amounts, it must indicate that something is wrong with spleen. If spleen is malfunctioning , then they are not removed &they remain in circulation.

DISEASES ASSOCIATED WITH HOWEL JOLLY BODIES   Splenectomy Hyposplenism -this can occur either from direct trauma or via autosplenectomy . Severe megaloblastic anemia Severe Hemolytic anemia  

2) HEINZ – BODIES They are formed by damage & denaturing to the hemoglobin component of RBC, most commonly by oxidative stress. The electron is transferred from hemoglobin to oxygen leading to formation of reactive oxygen species (ROS).The ROS led to damage of components of the cells e.g. hemes hence, Heinz bodies .

DISEASES ASSOCIATED WITH HEINZ BODIES Glucose-6-phosphate dehydrogenase deficiency (G6PD) NADPH Chronic liver diseases Alpha Thalassemia

3) PAPPEINHEIMER BODIES These are basophilic erythrocytic inclusions that are usually located at the periphery of cell . They contain iron and stains with Prussian blue They are pure iron deposits They can be confused with late reticulocytes They can cause false elevation of platelets count when performed with electronic counters  

DISEASES ASSOCIATED WITH PAPPENHEIMER Splenectomy Hemolytic anemia Sideroblastic anemia Megaloblastics anemia Hemoglobinopathius   The exact cause of this inclusion is not unknown.  

4) BASOPHILLIC STIPPLING   This condition is associated with fine and coarse granules located at in a fairly evenly distributed manner . They are the accumulation of RNA and Ribosomes   DISEASES ASSOCIATED WITH BASOPHILLIC STIPPLING   For fine granules-there are increased red blood cells production( polychromaticphillia ) For coarse granules-there is megaloblastic anemia, lead poisoning and thalassemia    

5)CABOT RING BODIES    They are literally loops, rings or figure 8 type structures located within cytoplasm of RBC They are quite rare They are micro tubular remnants of mitotic tubules that are involved in mitosis Their presence indicate abnormality in production of RBC   DISEASES ASSOCIATED WITH CABOT RINGS Myelodysplastic syndrome Megaloblastic anemia
