SKIN SNIP TECHNIQUE The gold standard test for the diagnosis of onchocerciasis remains the skin snip biopsy. PRINCIPLE A slide technique can be used where a skin snip is immersed in saline on a slide and covered with a cover slip.
PROCEDURES Wear gloves. Put one drop of Normal Saline on a clean slide. Add pieces of skin on the drop o f normal saline. Tease the tissue in small pieces under dissecting miscoscope . Cover with a cover glass. Let the mount stand for 30 minutes. Examine microscopically for motility by using x10 objective.
Report as positive for Onchocerca Volvulus microfilaria seen.
THE REPORTING OF RESULTS Report type of specimen examined and presence or absence of haemoparasites Example: Trypanosome and microfilaria. Indicate the date and name of technical staff reporting.
NOTE ON THE METHOD If then results are negative - Mount to stand for 4hrs and examine. If a dissecting microscope is not available: - Use table magnifying lens - Naked eyes All specimens must be regarded as potentially infections.
