UNIPORTER CATALYSED TRANSPORTER The plasma membrane of various tissues contains uniporter proteins that can transport amino acids, sugars and other small molecules. Uniporter carrier proteins catalyzes facilitated diffusion that is they transport molecules against their concentration gradient.

Uniporter is a type of protein that allows only single or one type of molecule to pass through them . They posses specific binding site for molecules and can undergo conformational changes upon binding with molecules.

GLUT 1 TRANSPORTERS GLUT TRANSPORTERS These are carrier proteins .There are various types of GLUT transporters present in human tissues that transport specific type of molecules depending on their location. GLUT 1 TRANSPORTERS Glut 1 transporter is a carrier protein encoded by SLC2A1 gene and belongs to the members of major facilitator super family (MFS)membrane proteins. It is expressed in fetal tissues, highly in erythrocytes and endothelial cells of blood brain barrier. they regulate the uptake of glucose in the cells and tissues they preset.

Glut1 is made up of 492 amino acids, That is 12 membrane spanning alpha helices each containing of 20 different amino acids .

MECHANISM GLUT 1 protein alternates in to two different conformational changes which facilitates the transport of glucose across the membrane.

When the binding site of the GLUT 1 transporter is facing the exterior of the cell (T1 STATE) ,the substrate molecule (glucose) binds to the binding site of the GLUT 1 protein. The GLUT1 protein undergoes conformational change and the binding site now faces the cytosol of the cell(T2 STATE). Where the glucose molecule is released into the cytosol of the cell. the opposite can happen when there is less glucose outside the cell. Inhibitors like fastenin,bay 76 inhibits glucose uptake

FUNCTIONS GLUT 1 proteins are highly expressed in erythrocytes plasma membrane for carrying the glucose in the blood. it is present in endothelial cells of blood brain barrier as it plays important role in uptake of glucose in cerebrum which is essential for brain as it is dependant on glucose as fuel source. it is present in fetal tissues and plays an important role in the development of pre-implantation of embryo.
