Object:- To Study the Various Common Objects. APPRATUS REQUIRED :- Compound Microscope, Glass Slide and Cover Slip MATERIAL REQUIRED :- Crushed Potato & Rice ( Starch Granules), Cotton F ibers, Woolen Fibers, Human Hair, Dust particles, Milk , Leishman’s Stain.
AIM The Purpose of this Experiment is to Familiarize the Students With Proper use of Microscope in different powers of objectives. Preparation of slides Take Clean & Grease Free Glass Slide. Place one Drop of Water on Each Slide and Add Different common Objects and Cover it with Cover Slip .
Identification of Different Common Objects Under low power microscope (10x) Under high power microscope (45x) Under oil immersion (100x )
Starch granules :- Take one Drop of Crushed Potato Solution on one Slide and rice on other slide. Covers them with Cover Slip and Observe it Under Low Power objective and than under High Power objective of Microscope .
Observation :- Potato granules are oval in shape in variable size with few concentric rings around a point called Hilum. Rice granules are polygonal and hexagonal in shape with central hilum surrounding by concentric rings. Each Granules Turns Violet when a Drop of iodine is Added to it .
Cotton fibers It Appears Thick and Twisted in Appearance Mostly White in Color. Undyed Cotton Fibers are Appears as Long Ribbon like Semi-Transparent Filament Which are Twisted at intervals. Two Faint Lines Appears Enclosing of Central Light Zone no Cortex & Medulla is Present .
Human hair Hairs are the Growth of Epidermis and Composed of Dead Keratinized Cells. It is Long Cylindrical, Filament in Shape. They Consist of three Layers – Cortex , Medulla and Outermost is Cuticle . Observation:- At high Power two Zones Cortex & Medulla is Seen, Outer Cortex is Thinner Zone and Black in Color and Inner Thicker Zone Contains Pigment Granules and air Spaces.
Woolen Fibers These are the Body Hair of Sheep, Rabbits and other Animals. They Appears as Long, Filamentous Structure and Cortex & Medulla. Fiber is formed and curly in nature so these Fibers Have Many Crisscross Patterns see in Medulla .
A ir B ubbles P ut a very small drop of water on slide and just drop a cover slip on it. Due to trapping of water air bubble are formed. It may be any size or shape. The single air bubble is rounded in shape due to its surface tension . They appears Dark & Thick Boundaries and Central Portion is Very Less Proportion. They are Refractive in Nature when Focus is Changed.
Fat Globules Put a Drop of a Milk and few Drop of Water on Slide or Take D iluted M ilk D irectly on Slide. Fat Globule A ppear C enter and Single W all.
Dust Particles Take Some Amount of Sand Particles in the Center of Slide. Put a Drop of Water and Cover it with a Cover Slip on Examination sand Particles A ppears as Yellowish B rown in Color and in Variable S hape & Size having Distinct B oundaries. Some Carbon P articles may be Seen in Between the Dust P articles. They are Smaller in Size and Found in Groups. We may also found some E pithelial C ells and fibers.
Stain Particles Take a Drop of Leishman's S tain on Slide and Spread it. Let it Dry at Room T emperature and Examine it Under low Power M icroscope and then Put a Drop of Cedar W ood oil and Examine U nder oil Immersion. The Stain P recipitate A ppears R ound, Uniformly D ark B lue V iolet G ranules of Uniform S ize Usually L ies S ingly and do not Forms C lusters.
