Principle: In FTIR analysis, Infrared light from the light source passing through a M ichelson interferometer along the optical path. The M ichelson interferometer comprises a beam splitter, moving mirror and fixed mirror. The light beam split into two by the beam splitter is reflected from the moving mirror and fixed mirror, before being recombined by beam splitter. As the moving mirror makes reciprocating movement the optical path different to the fixed mirror changed such that the path difference change with time. The light beam are combined in M ichelson interferometer to produce interference light. The intensity of the interference is recorded in an I nterferogram with optical path difference recorded along the horizontal axis. Applications: Identification of organic compounds since compounds will not have same IR Spectrum( by measuring absorptions). Qualitative determination of functional group predicted by present/absence of absorption bands. Quantitative analysis: by measuring intensity of absorption bands using Beer-Lambert law. Identification of impurities in drugs sample by giving additional peaks compare to the pure compounds. Department Of Pharmaceutical Chemistry,KLE CollegeOf Pharmacy, Belgavi. 15
