Title : To prepare slides of scale pattern of human hair. (E) Dr. Nutan D. Rathod Head, Department of Zoology, KCE’s Moolji Jaitha (Autonomous) College, Jalgaon
Objective To prepare a slide of human hair To observe and identify the scale pattern of the hair under a microscope
Materials Required Clean glass slide Cover slip Forceps Scissors Human hair strand Nail polish / clear adhesive Compound microscope
Procedure Take a clean glass slide Apply a thin layer of clear nail polish on the slide Place a strand of human hair over the wet polish Allow it to dry completely Gently remove the hair using forceps Observe the impression left on the slide under the microscope
Observations The impression of the cuticle scales can be seen The scale pattern appears similar to overlapping roof tiles Pattern observed: Imbricate scale pattern
Types of Hair Scale Patterns Imbricate: Overlapping like roof tiles (common in humans) Coronal: Crown-like (in small rodents and bats) Spinous : Petal-like (in cats, seals, etc.)
Conclusion Human hair shows an imbricate scale pattern This pattern is useful in forensic identification Cuticle patterns vary between species
Applications Used in forensic science to identify species Helps in crime scene investigations Can distinguish human vs animal hair
Precautions Use clean equipment to avoid contamination Let the nail polish dry fully before removing hair Handle slides and microscope carefully
